Antiproliferative Activity and Apoptotic Potential of Curcumin on Human Acute Myeloid Leukemia Cell Line (HL-60)

Curcuma Longa or also known as turmeric is a prominent natural remedy in Asia that has been utilized to heal diseases for decades. It demonstrated have substantial biological activities, including anticarcinogenic, antimutagenic, antioxidant, anti-inflammatory, anti-infectious, hypocholesterolemic, and chemopreventive properties. Curcumin therapeutic substance extracted from thought anti-cancer properties against variety of cancer types. However, its potential on acute myeloid leukemia (AML) remains uncertain [1,2]. 
 The objective this intervention study was examine the anti-proliferative apoptotic effects curcumin HL-60 cell line leukemia. A stock solution pure compound obtained by dissolving it dimethyl sulfide (DMSO). cells were treated with six different concentrations which 100, 50, 25, 12.5, 6.25, 3.125 µg/ml incubated at three distinct incubation period (24, 48 72 hours). vitro viability determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay quantified microplate reader 570 nm. Acridine orange (AO) propidium iodide (PI) staining employed evaluate morphological changes. MTT assays results show high cytotoxicity effect cells. maximal half inhibitory concentration (IC50) value consistently decreased when time increased. IC50 reported be 13, 8, 3.5 24, hours incubation, respectively. There significantly (p<0.05) between periods (Figure 1). alteration shown AO/PI appearance early apoptosis, late necrotic cells.
 These findings illustrated revealed an effective action human line. Further research required determine mechanism death compound.
